چکیده

BackgroundVery-early-onset inflammatory bowel disease (VEOIBD) is a chronic of the gastrointestinal tract occurring during infancy or early childhood. NOD-like receptor family, pyrin domain containing 3 (NLRP3) inflammasome has emerged as crucial regulator intestinal homeostasis; however, whether NLRP3 variants may modify VEOIBD risk unknown.ObjectiveWe sought to investigate and how rare variant, found in patients with symptoms, contributes development.MethodsWhole-exome sequencing bioinformatic analysis were performed screen disease-associated from cohort children VEOIBD. Inflammasome activation was determined reconstituted HEK293T human embryonic kidney cells components, doxycycline-inducible macrophages, well PBMCs biopsies variants. Pathogenesis using dextran sulfate sodium–induced acute colitis model.ResultsWe identified dominant gain-of-function missense variant NLRP3, encoded by rs772009059 (R779C), symptoms. Functional revealed that R779C increased pyroptosis macrophages. This mediated enhanced deubiquitination via binding deubiquitinases BRCC3 JOSD2, which are highly expressed myeloid cells. In model, NLRP3-R779C hematopoietic resulted more severe colitis, can be ameliorated knockdown JOSD2.ConclusionsBRCC3 JOSD2 mediate deubiquitination, promotes developing Very-early-onset unknown. We development. Whole-exome model. JOSD2.

Linkage of Parkinson’s disease in two very early onset siblings to a locus on chromosome 1

Parkinson’s disease (PD) is a prevalent neurodegenerative disease that usually affects individuals over 50 years of age. Age at onset in a small subset of PD cases is considerably lower, and these are considered early-onset PD (EOPD) patients. Most PD cases appear sporadic, but approximately 15% are familial, and some of the familial cases exhibit Mendelian inheritance. Genetic analysis of fami...

Late-onset inflammatory bowel disease in the very elderly.

Elderly-onset inflammatory bowel disease (IBD) will be seen more frequently with the ageing of the population and the increasing incidence of IBD. However, knowledge regarding the best approach to IBD in this population is scarce. Furthermore, differential diagnosis in elderly patients with diarrhoea, rectal blood loss or a changed defecation pattern is comprehensive and IBD is not always consi...

Maintaining Intestinal Health: The Genetics and Immunology of Very Early Onset Inflammatory Bowel Disease

Inflammatory bowel disease (IBD) is a multifactoral disease caused by dysregulated immune responses to commensal or pathogenic microbes in the intestine, resulting in chronic intestinal inflammation. An emerging population of patients with IBD occurring before the age of 5 represent a unique form of disease, termed Very Early Onset (VEO)-IBD, which is phenotypically- and genetically-distinct fr...

Defects in NADPH Oxidase Genes NOX1 and DUOX2 in Very Early Onset Inflammatory Bowel Disease

BACKGROUND & AIMS Defects in intestinal innate defense systems predispose patients to inflammatory bowel disease (IBD). Reactive oxygen species (ROS) generated by nicotinamide-adenine dinucleotide phosphate (NADPH) oxidases in the mucosal barrier maintain gut homeostasis and defend against pathogenic attack. We hypothesized that molecular genetic defects in intestinal NADPH oxidases might be pr...

IL-10 and IL-10 Receptor Mutations in Very Early Onset Inflammatory Bowel Disease

Very early onset inflammatory bowel disease (VEO-IBD) is a unique disease entity with a complex genetic susceptibility in affected patients. Next-generation gene sequencing techniques have revealed various monogenetic mutations contributing to the pathogenesis of VEO-IBD, including interleukin 10 (IL-10) and IL-10 receptor (IL-10R) mutations. In this article, we reviewed the features of and eff...
